Not if the yin seal is closed off, it won't. Gentle Fist can prevent Tsunde from accessing her healing abilities or even her CES.

That's not happening, since Rotation will simply rip her arms off if she tries to punch it directly, leaving her wide open. Even if she were able to heal her arms, it would take some time, which would be more than enough to finish her.
