Unfortunately for Tsunade, she lacks the close range combat skills that she needs to take out Kisame and severely pressure him. A punch would harm the swordsmen, but getting too close to him means that her chakra will be stolen at terrible amounts for her.

Katsuyu isn't nearly that large of a threat to summon who can quickly spawn lakes to to either push back the acid, or use the water to slow it down just enough for Kisame to evade.

A thousand feeding sharks is a jutsu that would most likely prove to be enough for the slug; that is if Tsunade's chakra isn't drained to the point where Katsuyu has a hard time remaining there. Which is a possibility. In short; once Kisame spawns a lake to flood the region, Tsunade is done for. At that point Kisame has gained far too large of an advantage for her to pull off a victory.

Kisame wins with mid difficulty.Ranshinsho would be Tsunade's only chance to defeat her opponent.
Restrict the water dome and it will be a match.

Tsunade is one of the weakest kages
she's slow and has no long range attack...
"one punch from Tsunade and it's over" her fanboys say
yet she failed to kill someone who wasn't even top20 and couldn't use most of his jutsus
dunno if all the sannins are equal or not but they are clearly overrated to the level that it's not even funny (except Jiraiya, thought those people who think he can beat Itachi because of that part1 statement are still annoying)
