I've heavily edited it and added more Manga Pages and stuff to give a stronger support to my Theory....
The Hidden Leaf Village was formed as a result of the Senju - Uchiha Alliance....
Before The Uchiha Clan Massacre by Itachi there were a lot of Uchiha in the Leaf Village but....
How many Senjus have been mentioned in the series upto now apart from Hashirama and Tobirama..??
I mean if the Alliance between Senju and Uchiha was the reason for the formation of the Leaf Village.... then where are the other Senjus..??
The only two Senju descendants in the series i.e. Tsunade and Nawaki.... Even they are not full Senju cause their Grandmother Mito was an Uzumaki....
If Madara wanted Peace in his childhood then why the hell he attacked Konoha using Kurama..??
And when did Madara really took Izuna's eyes..??
And when and why Izuna died..??
These questions give rise to the possibility that AFTER the Senju - Uchiha Alliance was made and The Leaf Village was formed The Senjus were Slaughtered and Izuna had something to do with it....
Now lets have a look at these two pics.... It's like in these two pics Kishi is hinting us that Madara took Izuna's eyes AFTER IZUNA DIED....
When you look at Izuna's body.... His eyes are covered with bandage which means his eyes were taken from the corpse just before his burial.... The Bandages may be to cover the marks caused while taking out his eyes from his corpse....
Here Madara his saying "AFTER My Brother died.... he only left me his eyes and their power...."
It means that Madara took Izuna's eyes after Izuna died....
Now Izuna was alive during the formation of Konoha.... See this image first....
It is the fight of the Valley of the end and Madara has EMS....
Madara tells Hashirama that he's different from the last time they fought as foes in a life and death battle....
And the last time would surely be before the formation of Konoha.... He said he's different because he had EMS during Valley of the end fight which means Izuna was alive during Konoha's formation....
Now a manga page from Chapter 619....
In the above image we see Tobirama telling Sasuke about Uchiha Love.... And how the loss of a loved one gives rise to hatred and darkness in an Uchiha's heart....
But....
Then Tobirama says something interesting - "The further down the path they fall.... The more powerful their eyes become and the harder it is to reason with them.... as in Madara's case."
And interstingly Hashirama tells Sasuke, "Madara's younger brother was his world and he loved his brother more than your brother(Itachi) loved you."
Just when Tobirama is talking that Madara couldn't be reasoned with.... Hashirama from nowhere speaks of his brother(Izuna).... Now that's weird....
It's like Hasirama remembered how Madara wouldn't reason with Hashi when Hashi spoke to him about what Izuna did....
But what might Izuna have done..??
I think after the Alliace between Senju and Uchiha was formed....
Izuna Slaughtered the Senjus and in turn was killed either by Hashirama or Tobirama.... And When Madara came to know about his younger brothers death.... he got mad....
When Madara asked the other Uchiha to support him against Hashirama and Tobirama.... The Uchiha shunned him knowing it would be foolish to clash with Hashirama and Tobirama....
They had achieved peace because of the Senju - Uchiha Alliance and didn't wanted that peace to disrupt and to see bloodshed again just because Izuna did wrong and got killed and now Madara wanted revenge from Hashirama and Tobirama....
Also I think that after seeing Uchiha support them instead of Madara.... Tobirama made the Uchiha incharge of the Konoha Police Force in order to keep that support intact.... He might have thought it best to show the other Uchihas that he believed in them despite of Izuna and Madara's breach of Alliance....
As said by Hashirama.... Madara loved Izuna above all and couldn't be reasoed with when he was told about Izuna slaughtering the Senju.... He was not supported by other Uchiha and started hating them too...

When Hashirama tells Madara that his idea of revenge is a slap to their brothers and Nakama(nakama is a japanese word for comrades,fellow country/clansmen).... Madara gets angry and tells Hashirama not to dare talk about love of comrades cause his brother was killed by Hashirama/Tobirama whom Madara considered a comrade and the Uchiha for whom he and his brother fought all their lives shunned him when he needed them most....
Also in the Latest Chapter 623 we see that Tobirama and Izuna know each other which means they have fought each other earlier.... Both seem to be their Daddy's boys who will fight each other on their Dads orders....
My Conclusion
Izuna who was full of hatred killed all the Senju after the formation of Konoha and was killed in turn by Hashirama/Tobirama.... When Madara heard of this he got full of hatred against Hashirama/Tobirama and even The Uchiha supported Hashirama cause they knew Izuna was wrong in breaching the Senju - Uchiha Alliance....
This further fueled Madara's hatred and he left Konoha to come back one day to get revenge and destroy Kohoha....
I know it's a bit too long....
