I thought the first time it was a mistake but you mention multiple times Senju Tobirama doesnt have Senju DNA.
On another note, I think its a combination.
Ootsotsuki descendant(Senju/Hagoromo or Hamura/Uzumaki) + Will of Fire is necessary.
Things like Sage Mode etc. are learned through training, they aren't necessary.
Hashirama was maybe born with mutated chakra and got Mokuton from it instead of just Water or Earth.
