II That chackra is what she used to Fertilize/Stimulate her enzymes (Enzymes are molecules; if Tsunade can fertilize them she heals on a molecular level)
III after she used that chackra and stimulates the production of her enzymes it accelerates her cells duplication rate, in which if she cant fertilize her enzymes in the first place she cant change or birth new cells. So her enzymes/large molecules are the essential things for her regenerating HENCE TSUNADE HEALS ON A MOLECULAR SCALE, NOT CELLULAR

Tsunade heals from Jinton whether you like it or not, it will pulverize her first sets of cells by the time those cells have been destroyed they have already been replicated and replace for another attack, as she said its her Tank of chackra that changes her Enzymes rapidly to birth new cells..
What i can agree with you is if, Onoki is capable to execute enough jinton to wear Tsunade out of chackra then and only then she can be killed by the attack, However as you can clearly see, it was STATED TO CONSUMED A LOT OF STAMINA BEING A KEKE TOTA, 3 jinton shots would clean him out and if he's in his prime i assume 5 jinton attacks would tire him out.

she doesn't fertilize cells but her Enzymes which enhance the growth of her cells, without the fertilization of enzymes there can be no additional cells, its like climbing a ladder an atom is more essential than a molecule, if your atom is destroyed you cant have any molecule, if proton or electrons are destroyed there can be no atoms in a body..... If molecules(enzymes) are destroyed there can be no cells. If she can stimulate Molecules she heals on a Molecular level, its just as simple as that.

As we know it the flames would ignite her skin, but working slow as usual(all the times it has been cast on someone, it took considerable amount of time to burn via them)Tsunade's YIN SEAL would already stimulating her Enzymes (Enzymes are LARGE MOLECULES THAT ARE SMALLER THAN A CELL) to enhance her cell production speed, to regrow a new skin layer, similar to how Byakugou healed her from those flames. The black flames are different however it was stated that it rots away what its been cast on, when it finishes its duty then it goes aside to another object. Your still misunderstanding jinton, when her regen is on she's already birthing new ENZYMES to counter jinton (being an attack on a MOLECULAR SCALE)by the time she's engulf Tsunade would already counter jinton with her second layer of ENZYMES AND CELLS for another jinton attack again. Jinton will only turn her first layer to dust, hence why she said she cant be killed because its her chackra she used to fertilize her enzymes to ACCELERATE HER CELLS RAPID REFORMATION RATE...

Imagine this situation:
Tsunade fights onoki and gets fully hit by an jinton-attack.
Inside her body the cell reproduction becomes insanly fast so that wounds can close up really fast.
All the new cells produced are still inside of her body.
Now the jinton hits and in that moment everything, everything of tsunade (her whole body) is turned into dust at ones.
All the old cells along with all the newmade and unfinished cells get destroyed at ones.
There is nothing left. How should new cells be made now? they are made by cellreprodution but there are no cells anymore to reproduce.
She would be just gone no comeback.

Amaterasu wouldn't stop burning after the first layer of skin, if that was the case then Nagato or the Zetsus wouldn't have kept burning after their skin was burnt off.
When Sasuke used oral rebirth he escaped Amaterasu because it wasn't yet done with burning his skin. Tsunade would just keep burning for 7 days or until she runs out of chakra.
How would she even seal Amaterasu while she would be burning in agony? Sure Amaterasu has been shown to be slow, but Sasuke was still screaming in pain. Not only that, but the scans you posted don't even say it was the yin seal.
