In other words her duplication of cells serves as a substitution.
The Amaterasu will now eradicate her first sets of cells then disperses to another nearby target as its first task has been executed, which was disintegrating what it was cast on, which was her first sets of cells or top layer.
Now that new ones as served as a replacement, uchiha Itachi or Sasuke would need to apply more black flames on Tsunade to disintegrate her new layer and so the process perpetuate until she's out of chackra and collapses.

Now Tsunade can also store this external chackra force/source in her YIN SEAL (Don't be confused it’s a powerful sealing jutsu that also has healing purposes) just like what she did with DAN'S.....
We have seen black flames sealed once by a shinobi and we have seen how another shinobi blocks the black flames without it burning via his defense. Gaara of the sand was able to infuse his chackra in his sand to protect A foot from rotten, the chackra based sand was then left aside untouched by the black flames, this should at least give you guys a better understanding as to whatever it TOUCHES FIRST THAT'S ITS ORIGINAL DUTY THEN IT GOES ASIDE.....
