You provided no answer. Explain Itachi's reaction to the specific question? Kakashi is practically saying he has the MS and Itachi knows this. Kakashi himself is hinting his sight loss due to using the MS. Later on it is revealed he has the MS. You gain eyesight loss due to the constant straining of the eyes not some random 2 eye theory. That much should have been obvious for the simple fact their eyes taking damage in the first place.

Here is a big difference in how Itachi reacts to what Kakashi's hypothesis about Itachi's MS powers.

How could Kakashi have known it causes blindness when he hadn't even used it enough to suffer the mildest effects from it?

The reason Itachi straight up stared at Kakashi's eye is because he could see the MS in it, after Kakshis comment. Itachi never stares like that at anyone, and at the same moment Kakashis Sharingan is shown looking back? It's obvious Itachi could see it.

Only when an Uchiha awakens both the powers of his Mangekyou Sharingan is when his eyes start to deteriorate. The collective strain of both those powers is what causes the blindness.
