He was referring to who we use to think was Madara. At this point in the manga, everyone thought Tobi was Madara. Of course he said that Madara was immortal because everyone thought Tobi was Madara without actual proof except hearsay. Madara not only had the EMS but Hashirama's power and yet he still died...

EMS only grants you one additional jutsu and it stops the blinding progress. You would think that infinite inzanagi would have been mentioned has that jutsu by name but it wasn't and also if Madara had that then he would have beaten Hashirama instead of getting pawned

From what we know ther are two forms of izanagi, the incomplete form and the complete form. Danzo's was the incomplete form whilst tobi's was the complete. In both cases the lost an eye, but it was the sharigan they lost. We can speculate that tobi who im 99.9% sure being an uchiha an having hashriama's dna was able to use the complete form of izanagi whilst danzo couldnt. After all its said to be an uchiha technique although from what we have read it requires hashirama/senju dna.
Now if sasuke was to get hashirama's dna and use izanagi, im not sure he wouldnt go blind due to having the ems which so to speak stops blind less, so this counters the effect of izanagi if used by an ems user. Logically speaking i dont believe sasuke would go blind if he used it, but kishi can decide otherwise.

Another question since Nagato was given rinnegan and he also was not an uchiha ,he could not use izangai during the pein invasion .The real sage of six path could use izangai because he had the rinnegan consisting of uchiha sharingan in it.He did not lose his eyes sight after because of the EMS features

Izanagi is a rennigan technique.
The reason why the sharigan can use it with the help of senju dna is because its a combination of the uchiha + senju powers = sage powers (rennigan).
The rennigan allows the user to all 7 elements which consists of yin and yang. Its this same elements that allows the user to use izanagi. Nagato had it within him to use it without going blind, but maybe he hasnt mastered it or maybe we didnt get to see all of his techniques
