With full intel I think Tsunade can win this with a lot of struggle since he knows about all of their jutsus. But I'm not sure, it could go either way.

Also there are multiple threads where it's been proven that Tsunade would lose to Neji without Katsuyu. Due to the fact that there are all sorts of CQC fighters and long ranged fighters in this fight, the long range fighters can easily escape Tsunade's range.

Byakogou isn't tanking gentle fist, chakra loss from Shino's bugs or getting sliced to bits by Kiba or Temari. Pretty sure Sakura's punch would also incapacitate Tsunade but that is debatable. As for Katsuyu... well let us just say that its acid gets blown away by Temari right back at Tsunade. Furthermore, if Tsunade or Sakura do summon the slug, its huge shadow will give Shikamaru the ability to capture Tsunade and slow her down occasionally.

I'm going to have to say team 2 low difficulty.

Sakura basically stalemates Tsunade seeing as they have the same ability [Sakura having the edge in strength, Tsunade in healing], bringing in a team makes this a stomp.

nothing says she doesn't. I mean shizune learned all her poison from tsunade. Even sakura at one point walked with sleep bombs and poison. It wouldn't be out of place to say she walks with poison as well seeing how she drugged jman.

But if you want to make a real argument against it you can even say she doesn't walk with ninja tools seeing as how she has no pouches or packs to hold said items.
