Katsuyu summoning. The size should burst it and the feedback from the space time jutsu should dispress the water. And If she really want to get down a dirty she can liquidfy and Tsunade can kill him with medical ninjutsu like she did with Orochimaru.
Yes he can absorb chakra, but medical chakra is different in nature.
Not to mention Katsuyu's acid in the water should kill him and his sharks, Tsunade would be fine because she either have immunity, or she'll just tank and regenerate from it.

Tsuande doesn't need chakra to kill him, and a plethora of her chakra is sealed away, she cant get it. He only got to naruto's in part one because naruto was entering the cloak mode.

Kisame outmaneuvered and overwhelmed base Gai and V1 chakra shroud Bee in CQC with relative ease. Tsunade, who is slower and less skilled then these two, will be having an awful time in CQC against Kisame. Especially on the surface of a large body of water at Kisame's disposal. Sharks attacking from below the surface in tandem with Kisame above the surface would overwhelm her.

I believe so. Therefore, she can escape Dome as katsuya divides into multiple mini clones, with one absorbing Tsuande.

Well that isn't very nice mister,
Tsunade > Base Gai Lol. She isn't slow, her fight with Madara proves this, with her blitz and rinnegan user, and taking down susanoo clones (while fighting sloppy). And Orochimaru's face is still recovering from being punched by a reduced strength, drunk and restless Tsunade. Tsunade fights by leaping a lot, and she hasn't really missed a target , and she faced speedy foes. She was even going to kick the Raikages ass for attacking Naruto, so speed isn't an issue with her. She prolly pick up a shark and throw it at kisame,
