Tsunade having been a medical ninja for many years can hear erratic heartbeats from a far (x). Kakuzu's heartbeat wont be erratic but he will have 5 different heart beats which will make enough noise for her to hear. At that point, Tsunade will know he some kind of monster and be on guard and wont fall for no surprise attacks.
Nice attempt but no, there are more ways then hearing to tell a erratic heartbeat. Perspiration and the dizziness and unawareness of his surroundings are also a likable give away for erratic heartbeat

Kakuzu wont mind taking a hit with his domu activated as he thinks he can take any physical hit (x)(x). But he has no clue on Tsunade's strength which will rip through Domu(x). 1 heart down already.

Neither Blunt Force, Nor Explosions will ever come close to even denting it, The only way known to even make a scratch is with a pinpoint laser or other diamond U_U Now this is for physical attacks of course obviously some jutsu will be able to easile break Domu, however a CES Punch? No Way. Not that it matters anyways Kakuzu has shown quick reaction and avoidance time in CQC Situations

Fuuton - Whether the wind will be slicing (x)(x) or blunt force pressure (x)(x). Its something Hidan survived at point blank range(x), he has shown no above average durability.
Raiton - Its said to have piercing power. (x)(x)
She can survive those abilities but she can also simply dodge them. Tsunade has shown great speed and power in making giant leaps.

Which is GG Once removed her healing is no way creating a new functioning heart repairing organs is one thing but fully growing a new one? Not possible with her cellular division alone.
Bringing out the masks is also a bad move, Tsunade can survive and dodge the attacks and one hit from her and the masks are done. They were easily killed by tenten (x) and Chouji (x)(x). Tsunade hits much harder than both the attacks.
Have you not read the manga? Ten-Ten Got lucky a misfire from a different battle entirely caught the heart off guard

Pulling would only spawn more threads yes it may nudge him forward a bit but the brunt of the force of the pull would be on the threads inside of Kakuzu....Once she's restrained she is a sitting duck for Kakuzu's heart removal.

She can simply cut the threads. There is no blade sharper than a chakra scalpel. It was able to cut Naruto's skin so sharply to the point where there was almost no blood. (x)

There is not indication the the mask are the elements themselves, they are made out of the threads, same as Kakuzu. All you have to do is destroy the threads, the Katon and Fuuton masks werent killed by an element that countered them, they were killed with an attack which damaged the threads. [

None of the mask can fly, its has wings yes but so do penguins, chickens and ducks. Yet none can fly. It most likely has great jumping ability.
