Did anyone else notice the diamond on both Mito and Tsunade's head ? Also Mito is Tsunade's grandmother so is it something she has passed down to Tsunade ?

She is part Uzumaki, yes. It's likely that the diamond thing was a technique passed down by Mito, or it could be Tsunade's unique jutsu that she formed in that way to honor her grandmother. Nobody knows.
