True, but if he was outnumbered, contstantly fighting running out low on chakra, possible even clashed with All First Kages then First Jinchurki were unleashed on him and maybe even attacked from behind ?

I am sure it would takes him out.

Another possibility is that he just run out chakra and then got poisoned by someone

Actually it makes a lot of sense that he wouldn't die from overusing his healing ability, I mean Obito took his cells and he was able to use his eye almost indefinitely (Kamue) without any visible blinding effects, But this was Harishama himself , I think he would be able to use it without killing himself not to mention that jinchuriki not having that much effect (due to not being perfect) thing Gerkak mentioned.
