Hello guys, I would like to talk about possible chain of actions which led to Hashirama's death. Please bare with me If it stretch too much. Here I go:
^^ Hashirama could heal instantly with no seals formed just like his grandaughter Tsunade could do in Byakogou seal.
There is known statement: "The greater power, The greater risk":
^^ As we can see when Tsunade describe her own ability known later as Byakogou while Shizune mention the risk of such incredible technique. The risk is shorting her own life, just Like it could in Hashirama's case.
Hashirama has fought so many battles so clearly it could shorting his life greatly.
^^Hashirama gave all Tailed beasts besides Kurama as sign of their treaties. First with Five great countries, later with other ones. It was way to stabilize The balance of power according to Hashirama.
^^ Elder Granny Chiyo explained how People tried to contain incredible power of Tailed beasts containing these so called ultimate chakra weapons inside human bodies making shinobi a jinchurki.
^^ The Five First Kages requested and even demanded to recieve Tailed beasts as good balance of power.
^^ First Kazekage suggested possibility of using Bijuu against other to gain his demand
Soon First Great Shinobi War occured which Hashirama wanted to stop by sharing Tailed beasts, but this is what wiki writes about it:
In short, after getting Tailed beasts The war took even worse direction as Kage created Jinchurki to contain Tailed beasts powers and use it for their need at whim.
^^ After taking look at Hiruzen's flashback where we saw Hashirama last time. It is not farfetched to say that Hashirama has to die in First Shinobi World War.
So in The end it was Hashirama overusing of Healing ability with no seals which keep shorting his life + Getting ganged up by all First Jinchurki at one time in First war. He may stop or deal with them, but after this battle He most likely die.
Here is another interesting remark how Hashirama reacted about Jinchurki word:
^^ Here Hashirama express his worry about Jinchurki word, not specific Juubi Jinchurki. Also in other translation he said: "Jinchurki... it is bad..."
Other two options are short:
- Black Zetsu poisoned him in sleep, doubt he could sneak unoticed when Tobirama is near especially
- Fight againt Madara weaken him so much that he was far from full strength and then maybe too much rengerating side effects + possible attacks of Kage,jinchurki killed him
Of course it could be any other reason, but it seems most likely. Also wouldn't it be irony how own power/abilities with no proper mind can kill even yourself.

Summary :
- First action which led Hashirama to death was overusing and too much depending on Healing ability with no seals which shorted his life
- Second action which led Hashirama to death was delivering Tailed beasts to other countries
- The result was getting ganged up by all first jinchurki(maybe poisoned eariler as well) Hashirama was Killed or left at verge of death after dealing with them. Soon later he passed away.
Of course it could be any other reason, but it seems most likely. Also wouldn't it be irony how own power/abilities with no proper mind can kill even yourself.
