I will try and make this as much like 'show and tell' as I possibly can for those who dont like to read essays. But before you start reading I must warn you, this quite possibly might be the final piece of the puzzle and a major spoiler! You have been warned!

Zetsu, White and Black, sort of have an undying loyalty to Tobi. Even more so than the Akatsuki members. You all probably know this by now, but he is Tobi's number one asset. Information gathering, tracking and sensing chakra are just some of his key traits.
But the most important thing that you must learn about Zetsu, White and Black, is that he is not a human being!
As this manga page proves, Zetsu was created from plants! White Zetsu's DNA is a carbon copy of Hashirama's DNA. Keep this in mind.
Tsunade, being the Granddaughter of Hashirama, is the only person in the world who knows everything about the properties of Senju cells.
And the above page clearly states what kind of use those cells have been put to.
We can deduce that:
1. Organic life forms like plants can be given a human form. Plants and trees can live hundreds of years.
2. They can be cultivated like crops, to create more of the same, multiple times.
3. Once Senju cells have been mass produced, the user of those cells can periodically administer them to himself over and over to a point where he can be considered an undying mortal.
This is key in understanding Tobi's claims to have been alive since before the beginning of the Hidden Villages and explains his vast knowledge of the Ninja world, particularly around the era of the First Hokage.
Most people treat Mokuton as just another Kekke Genkai. It is the most unique ability in the world, far superior to any other blood line limit, and here is why:
Shodai had the amazing ability to turn his chakra in to a source of life!
It is the closest thing to creationism in Naruto, Hashirama could literally give birth to entire forests with his chakra.
We all know what Tobi has hidden in his basement:
Apparently...that 'thing' is the result of the same infamous Senju cells that were stolen from Hashirama during his battle with Madara. Metaphorically, that is his bread and butter and he would be nowhere without it.
I believe it is now safe to assume that both the Zetsu were born from a successful experiment by Tobi, and in a sense you could say that he is their Father or creator. That would explain Zetsu's extreme loyalty to him.
Now we get to the fun part, you all know how Tobi shares some of the same traits as Zetsu, mainly the white substance that constructs parts of his body. He has been seen sacrificing and discarding limbs in battle without hesitation:
The important thing about that white substance is the fact that it is White, which means Tobi's body possesses the same traits as White Zetsu.
Here is something you might have overlooked before, and it is the key to everything, so much so, that it is almost unbelievable that Kishi would divulge such sensitive information, even if it is only a sentence:
Zetsu is revealed to be a type of parasite! A parasitic plant life form created by Tobi. Zetsu is able to duplicate and replicate any living being he comes in contact with, down to trace amounts of their chakra, now this is not something a parasite does, it is similar, but a real parasite, inhabits its victims body, lives in it, off of it....ultimately consuming whoever that person is.
You guys wanted to know the connection between Tobi and Zetsu....well here it is:
Tobi is the original parasitical life form created by Madara from the cells of Senju and his own. Consisting of possibly the strongest genes ever, he inhabits an Uchiha body, Obito's body. Reconstructing whatever limbs may have been lost, and by also having access to his memories and thoughts through the cerebral nerves in the brain, including his own, Tobi became the man we saw in Chapter 599.
Tobi is the original Zetsu!
