I was doing some reading about Yin & Yang Release and was also looking at a thread from someone who was mentioning something very interesting. I am sure it has been discussed before but am not sure. Sorry.
Anyways, as I read there was somethings I had to read several times to make sure I got it right. Yin is the concept of giving imagination a form and Yang is the concept of making imagination's form a reality.
In other words, Yin would be like a genjutsu giving the appearance form but not actually existing. Yang on the other hand, breathes life into form. It is the essence of reality. When you combine these two you can pretty much make concept into form into creation. A good example is Izanagi.
Now when the SO6P divided his powers he divided them using a Yin & Yang concept.
From what I know:
- Hashirama had superb control of this rare kekkei genkai.
- Yamato's Mokuton paled in comparison to Hashirama. (According to Yamato himself)
- Danzo's version significantly drained him of chakra so his use was limited.
- Interestingly, when Danzo's chakra levels dropped significantly he began to lose control
of his Mokuton. This probably hints to their being a considerable and ignored requirement
for being able to control it and manipulate it.
- Reminds me of a tale of a Spainaird who went to go look for the tree of life. When he found
it he was close to death so he drank form the tree. Within minutes he was restored to
health only to seconds later become a tree.
Anyways, my guess is that the Mokuton is reacting to the Yin chakra of the Mokuton. But there is something also weird. The "weird" effect has only happened with Zetsu's. We have yet to see how it reacts to Yamato. What did Yamato have that is so special and crucial to the evolution of the Zetsu?

The Older Son received the Yin half. With his dojutsu he was able to control the realm of concept into form. Which explains why the Sharingan/Byakugan/Rinnegan are naturally better at Genjutsu or being able to see/detect chakra flow including disturbances in its flow. They can control the perception of the senses using their dojutsu.

The Younger son received the Yang half. Yang breathes life into form. Which helps us understand the Senju's kekei genkai, Wood Release. Using this power they were able (at least Hashirama could) combine two elements, water & earth, and create life. The Yang half also allowed them to inherit vitality which would explain their rumored vast chakra pools, this includes the rumored long life of their distant cousins, the Uzumaki.

That said, the Yang chakra that is used for his Nine-Tails Chakra Mode has a strange effect on Mokuton (Wood Release). Now I wonder why it does that? Is it reacting to a special property within the Mokuton that we have yet to know about? From what I know:
