Proof? Suigetsu's DNA test proves nothing since we don't know whose DNA sample was used for the test. It could possibly be Sasuke's. How can you be so sure that is Karin's DNA sample?



You clearly have no idea how DNA testing works. Just touching something doesn't transfer your DNA to that object. It has to be someone's biological part to determine whose DNA it contains and then comparing.

The DNA sample Suigetsu used was probably Sasuke's, but Suigetsu thought it was Karin's. That's what led to this misconception.



Sharingan is awakened through emotions, and Sarada's emotions were completely genuine. The means causing those emotions were false. Sarada didn't know that.

I don't believe that Sarada is Karins child. Suigetsu probably screwed up and actually brought out Sasukes dna sample.
I mean come on...do you really think Sasuke would do it with Karin. Karin might have taken his semen from him and got busy, but I don't think sasuke would willingly (bang) Karin
Grow up and think outside the box.

Maybe Sakura couldn't have a baby because she's infertile and Karin decided to be a surrogate or something. This is a manga directed towards younger males, do you really think Kishi would have Sasuke cheating on Sakura or having a one night stand?

if Sarada was the experiment of Orochimaru that would honestly be a great writing maneuver on sensei Kishi's part. And these introductory 7 chapters have set that perfect parallel to jump off of. If this be the case, then hopefully whoever writes the next part skills it and rekindle the series back to prominence.

Can't wait to see DBZ

Nor are most entertainment productions, for that matter.

Assuming Kishimoto sat down and did some research, he would know that the best means of determining maternal lineage is through mitochondrial DNA, since all of our mitochondria come from our mothers. Thus, direct maternal lineage can be obtained through an analysis of mitochondrial DNA. Determining paternity of a female is not as simple and has a much higher margin of error since one has to look at nucleic DNA and infer how many markers were likely inherited from the father. Generally, paternity tests of females is best done to eliminate potential candidates. Paternity of males is easy to establish since the y chromosome is inherited directly from the father and is, actually, a very short chromosome that is easy to track mutagenic changes through.

Anyway - if Suigetsu's machine works off of mitochondrial DNA (which is stable enough to perform an analysis after several years and can be obtained from hair other than the root), then the only scenario in which there could be a match between Sasuke and Sarada is if Sasuke's mother is from the same direct maternal lineage as Sarada's mother. IE - Sarada's mother would have to be the daughter of a sister or maternally-linked cousin to Sasuke's mother.

If this is how his machine works, and this is how Kishimoto envisioned it, then it is most likely the case that Karin is the only logical match. Granted, the cited time of 1000 years is not great enough to produce substantial mutations in the mitochondrial DNA - so in theory, if the Uzumaki do not predate Kaguya (I suspect they do, and that the Sage's father was an Uzumaki), then it is logical to assume that women of Uzumaki, Senju, Uchiha, and Hyuuga are still part of the same maternal tribe and would all appear as very similar matches. Thus - if Sakura traces her maternal lineage back to Senju, Uzumaki, Uchiha, or Hyuuga, then she could theoretically appear very similar to Karin from a mitochondrial standpoint.

Further, Sasuke would appear very similar to Karin under such a context, as well, assuming both have a direct maternal lineage tracing back to Kaguya.

The only other way a match could be obtained from Sasuke is if one is going off of nucleic DNA. It is not possible to get this from strands of hair that do not include the root, and it is difficult to get quality samples. While flakes of skin, sweat, and saliva can contain DNA, getting clean samples from these sources for analysis is problematic. Rarely do you get large portions of the genetic code, and you often have to introduce various polymerase strategies that are known to be the source of errors when identifying strains of viral infection (often inducting other strands of code from the host's destroyed cells into the chain).

Contamination from various other DNA sources is, therefor, an issue with these types of samples. If rubbing one person's face on a shirt is enough to leave behind DNA that can be sampled - then the person who was wearing it first also left behind substantial traces. This is why these types of samples are generally used only as a last resort and must be given a wide margin of error even in the case of using them to positively identify an individual - much less use them to infer maternity.

But, we're dealing with Naruto, here.

The only thing we can assume for the moment is that the machine is accurate in its analysis - whatever sample was placed in it has correctly identified a match to Sarada as being from her mother (the machine is not mistaken or flawed).

This leaves two possible scenarios for this to point to something other than the direct interpretation:

As the OP states, that the sample is somehow not from Karin and is, instead, from Sasuke (thus, the machine has identified a match to the father).

Or that Sakura and Karin have a very strong or close common ancestry.

I suppose a third option does exist where Sarada is some kind of experiment from Orochimaru who Sakura opted to take care of. Or maybe an experiment from Karin...

Or... perhaps Sakura is Karin in disguise...

The real Sakura is out there, somewhere (though one would think that Naruto and others would have noticed the difference... but...).

I have no particular preference for any of the outcomes.

Further "look underneath the underneath" is incredibly vague.

My argument that Sakura and Karin could be of very similar ancestral origin and therefor indicate a match is "beneath the underneath."

I was correcting you on your use of NCI to gain ideas about how genetic sequencing/comparison works. NCI essentially treats DNA as a magic fingerprint that you leave anywhere. Whether or not that idea is relevant to Naruto's universe is yet to be seen.

From an author's standpoint:

Kishimoto has chosen the theme of parenthood - what makes a parent and what makes a child. The introduction of the idea that Sarada has been raised by Sakura but is Karin's child is a solid core conflict to use to explore this theme, especially since Sarada says that Sakura "means nothing to me."

Thus, the story will likely play out with Sarada coming to an understanding that being a parent is more than simply genetic lineage and heritage. Ideas and personality are passed on - just as Naruto mentions that Sarada's personality/aura/energy is similar to Sakura's.

This is simply a more dramatic way of depicting the old 'adopted child' story where the adopted child sets off to learn about his/her 'real' parents to find out that they received more from their adoptive parents than they initially realized or appreciated. There is also the opportunity to introduce some twists in this idea - that people aren't as different from each other as we may suspect, that we all share some of the same origins, and that children can just be paranoid little shits.

I've no idea what Naruto's left arm being made from the cells of the first hokage has to do with this specific issue, unless you are trying to insist that Orochimaru was able to extract cells from a guy who died either a few centuries ago or last decade, depending upon which part of the manga we are reading.

Yamato is supposedly a project of Orochimaru who has Hashirama's abilities (it isn't clear if he's a type of clone, or a genetic splice, or what, since he doesn't resemble an identical clone). Either way - various interests have kept a culture of Hashirama's cells around for some time. It isn't clear exactly where the cell lines came from or how they've been passed around. I look at them to be more of a "green rock" for the series.

I believe Karin with Oro's help artificially inseminated herself with Sasuke's juices to have his child. Now the big question for me is why if Karin is the birth mother would she give up her child to Sasuke's wife to raise? It all smells of Soap opera shenanigans to me but i'll sit back sip my ice tea and wait.
