If you train to unlock your sharingan, and get to two tomoes or three tomoes, whatever, and then drop the bio, do you have to go through the training again?

No it stays with you when you make another uchiha bio

Nope. All of your training goes to any of your bios in the future as well.
So, any jutsu/kekke genkai you learn will stay with you forever.
