Well that isn't the way that she acts when Naruto questions her about her chakra levels after she uses it. Had it been previously stored chakra no one would have worried about Tsunade being out of chakra.

Either way I think the gates could only be used efficiently by either Naruto or someone with insane chakra levels and Samehada (Samehada heals people with chakra)

Samehada doesn't heal anybody, it's not a medical shinobi, is a SWORD, Samehada restores chakra that takes from enemys , tHATS ALL.
Tsunade got run out of chakra when Pain attacked Konoha just because she spent all her chakra healing ppl throughout Katsuyu and finally she had to release her forehead seal to use her HUGE HUGE HUGE amount of sealed chakra to use Creation Rebirth over ALL KONOHA,,, I mean 10.000 ppl or more.
Tsunade's levels of chakra are considered Bijuslike when she releases the seal.
Plz!!!! I'm begging you, read and reread Naruto's Mangas, Naruto's Database, Naruto's Wiki and watch carefully Naruto's Animes.... and once you think you have understood something,,, come back here and try to make some real arguments,
