Okay so this weeks chapter gave us new insight into how Doffy and Luffy match up and for the first time since probably Rob Lucci, Luffy has come up against a foe who is physically as strong or stronger than him. If you look at some of his past opponents, it could be said that the likes of Crocodile, Enel, Magellan, Smoker etc who all gave him a rough time were weaker than Luffy in terms of pure physical strength (disregarding abilities) but held the advantage over him because they has fearsome abilities such as Crocodile's sand manipulation, Magellan's poison and Smoker's intangibility.
However, in comes Doffy. I've seen many people on the base claiming that Doffy's fruit is a bad match up for Luffy and he has shown no ways of blocking it. Yet even then, if Doflamingo was to fight Luffy purely with his physical abilities (punches/kicks etc), from what we've seen this chapter, Doflamingo would still mid dif Luffy at best.
Doffy took one of Luffy's strongest attacks, without arming himself with buso haki and was able to get up without any problems. The middle left panel shows that Luffy was in gear second when he did the Red Hawk so it should be on a different level to the attack that hit Hody Jones and yet Doffy shrugged it off like it was nothing.
This panel shows Doffy's haki matching up against Luffy's. It's a clear win for Doffy showing that in terms of pure physical fighting prowess, Luffy can't beat Doffy, never mind Doffy's strings.
So basically without a powerup or the activation of some yet unseen ability, Luffy has no hope of even challenging Doflamingo on a physical level and it's likely that even Luffy and Law together as Luffy is now couldn't beat Doflamingo because of the vast difference in physical ability.
