Two or a sannin is enough. I think Itachi wil probably lose to Tsunade. She may have the least ammount of damage potential (slash the punching raw strength part since its melee) but she is the most durable character in the Naruto universe. She is definitely immortal and with the latest manga chapters her technique "Strength of a hundred/ thousand men" really enphasizes that she is immortal... She wont die ever. And what happens if Amaterasu fails to kill her, then she gives a lethal punch. a single is needed...

Oh and since this is three to one, Itachi Edo is soo gonna lose...

Blaze Release is not a God. He is a person who is rather eloquent and uses his words in a convincing way. But as the sophists proved long ago, Just because it sounds plausible it doesn't necessarily make it true.
He's not taking Jiraiya Mid-Difficulty. Two sannin would overload him. Three Sannin is way too much.
