Not really.
Jiraiya never had chakra problems, and his loss to Pain wasn't due to chakra depletion; it was due to flat out inferiority. People like Naruto, Jiraiya, Hashirama, Tsunade, Kisame . . . it doesn't matter much whether or not they're Edo because they had tons of chakra in life anyway, only suffering from depletion in severe situations.
