Tsunade most likely met Katsuyu though Mito or Hashirama. (1) Mito had the Byakugou seal which is directly linked to Katsuyu (2) there's a good chance that Hashirama learned his variation of SM in The Shikkotsu Forrest (Katsuyu's residence7).

I'd say meeting Katsuyu through Mito is more believable.

In this instance Katsuya's ability to heal the allied shinobi is proportionate to the amount of chakra built up in the seal. Makes sense given how medical ninjutsu works. The more chakra stored in the seal, the more chakra that can be distributed. Simple supply and demand. Doesn't mean that Tsunade learned her seal from the slugs...just means they can be used in tandem.
