Tsunade can easily catch Sasuke off guard like she did with Orochimaru and Madara. If he doesnt know about her regeneration, he can stab her in the heart and think she is dead then she can easily hit him.

Then Sharingan sees her movements and his wings block her hit. She won't be dead until her chakra is completely gone, and Sharingan can see chakra. So why would he think she's dead? Lol. She never caught Madara off guard in the way you are describing. In fact he reacted to her surprise attack accordingly.

CS2 wings cant last all day as the curse mark damages the user.

Doesn't need them all day as Tsunade won't last that long.
