This doesn't mean that they all are dead, it simply means that at least one of them is dead. He may have 1,2 or maybe 3 brothers still alive.

Izuna was alive when they are adult and Madara leading the Uchiha Clan so, at least Izuna is alive that point but i don't know other 3 of them alive or death. But it's certain, He lost 1 of them at least, it's why he knew that feelings
