*I grip the handle of the bokuto with both hands. I then raise it and line up the blade with my shoulder, as though I'm preparing to cut downwards. I then place my legs apart, with my left leg slightly in front of my right, completing the stance.*

Like you said, the stance can be used to quickly switch to a needed stance. I myself would use this as a default stance. That way, instead of guessing what my opponent will do and attempting to go into a stance and hope I'm right, I can adequately switch to a needed stance.

It can also be used for the downwards strike.

Okay, so when you tilt the sword over your shoulder, your attack are quicker and more deceptive but have less range and strength. Depending upon the angle and prior action, your stance can appear as if the weapon is held behind the head or neck - you can simply create this illusion by moving your waist. xd

So, let's begin with attacks.
What is the difference between an one-handed and a two handed-slash?

A two-handed slash is faster and stronger due to the fact that two hands produce more force than a one-handed slash.

But in what circumstances would you prefer to use an one-handed slash instead?

I honestly have no idea, stabbing techniques come to mind.

*I hold the blade with my dominant hand, my right hand. I hold the blade up and past my left shoulder diagonally, with the handle placed just in front of my left pectoral. I then slice forward and diagonally. As my strike advances, it starts going downwards rather than straight forward. The strike leaves a wound on the clone, starting from right shoulder and moving down his body, going in a diagonal angle across his left chest, abdomen and ending just above his left hip.*

I then bring the bokuto down in a straight line, slamming the wooden "blade" into the clone's shoulder. The force of the impact breaks its shoulders, causing it to vanish in a puff of smoke.*

Yeah. It's basically forcing the tip of the sword's blade into one point on an object. This can create a small wound, but can be very deadly.

*I grab my sword with one hand, gripping the handle tightly. I then walk up to the clone. I bend my elbow back, drawing the sword back as well. As I pull it back, I point the blade towards the clone. I then push the blade forward until it pierces the clone in the stomach. I pull the blade out and it disappears.*

Hmm. I'd say either at the beginning of a match where the opponent's defense might not be at its best, or at the end of a combo to finish them off.

Well, the surprise from a lunge could leave the enemy distracted, and break their concentration. This could be taken advantage of so that if the lunge hits a vital spot, the opponent can be defeated.
