Hashirama with 100% Senju body + Rinnegan (Sage Eyes) would have been the closest person to the Sage of Six Paths.

Sage mode Hashirama already defeated EMS Madara, so we have to assume that he has counters to what you listed.

Honestly there's no telling how the rinnegan could empower a full senju.. but we do know that without PS and born Uchiha blood his dojustu would be limited.. and it's unknown if a Senju could awaken the rinnegan that way..and if it did the chakra strain would be ridiculous for a non-uchiha

Nagato being an Uzumaki (close relative of the Senju) was well suited for the Rinnegan.
He was able to utilize its power to almost full potential, with no considerable chakra strain.

Rinnegan is not exclusive to the Uchihas so Senjus could unlock it too. All they need is the Uchiha eyes.
After all both are the direct descendant of the sage and rinnegan is the Sage's eyes.

It's obvious that Hashirama has knowledge of rinnegan as he have knowledge of the jubi itself. But unlike Madara, I doubt if he have the knowledge of how to acquire rinnegan.

I think Hashirama with Rinnegan could have solo'd all the other Shinobi nations with ease. Just like how Nagato solo'd Konoha.
He would have united the shinobi villages into 1 uniform country and thereby preventing the previous 3 other great Ninja wars.
Rin wouldn't have died, and the Amegakure trio (nagato, yahiko, and konan) wouldn't have suffered the consequences of war.

I think Hashirama had a chance to unite the shinobi villages, but he was too passive to do it.
On the other hand Tobirama would have done something about it for sure, if he had similar power remotely close to the Rinnegan.

It is pretty obvious how Madara unlocked it: You just need an Uchiha dna (via the sharingan) and Senju dna (via Hashi cells).
Since the sage was the ancestor of the Uchiha and Senju, in order to obtain the Sage's eyes (Rinnegan) you need both dna requirement to mimic that of the Sage.

I believe that it took Madara almost his lifetime to unlock the Rinnegan due to not having 100% Senju body (which acts like the catalyst to transform the Sharingan into Rinnegan).
With Hashirama it would have awaken with a shorter amount of time since his body is pure Senju
