On this chapter Kabuto and Itachi have a conversation, back then we couldnt understand what Kabuto was talking about but now its much more clear, read the bottom row and see how after what Kabuto says, Madara shows up, which implies that Kabuto was talking about Madara:

That means somehow Tobi knew what its power was. Im assuming he has a way of finding out the MS powers of each Sharingan, or maybe Tobi knew Obito is related to someone who has great Sharingan powers and he wanted these powers for himself. Its just an assumption, discuss.
