Might Guy vs Tsunade!

Tsunade. If guy goes any gate lover than 6th then she dosen't need to regenerate.If gu goes 6th or 7th gate then she will need to regenerate.

Gai would need to go 7 gates and even that, tsuande would survive due to her medic ninjutsu. Afterall gai's strongest attacks hasnt exactly killed anybody. If a weakened kisame survives somebody like tsuande should survive.

Once 7 gates takes its toll on gai, he would be alot slower for tsuande to give him a one hit ko

Enough chakra to heal hundreds of villagers in the pain invasion, and tank choushinra tensei....

Gai's strongest attack (afternoon tiger) fails to knock out kisame, a kisame who didn't try to dodge or even block, and took the attack right in the face. Gai has nothing in his arsenal that can spell instant death for tsunade, which is the only way he can kill her.

Remember tho, it takes time for gai to enter his gates, he puts his arms together and he names gate names and tsunade then activates it

Plus tsunade has plenty of chakra up there, think about it Tsunade protected the whole village with katsuya using CR while pain used Shira tensei to destroy it

She has crazy chakra up in her forehead

Tsunade wins with her regeneration jutsu which would make her pretty much immortal in battle, so even if gai is too fast she will regenerate also her incredible strength would be of good service.
