Enough hatred. Uchiha and Senju were killing each other for a long time prior to Konoha yet they still united anyway. Senju didn't directly kill Uchiha in Konoha. Hurizen, Danzo, Homura, and Koharu took things upon themselves. Hashirama would never allow such. There are 2 reasons why the Uchiha were ousted in Konoha.

1) Madara was misled by BZ
2) Uchiha clan wanted to be in constant war (with everyone and with themselves) due to the nature of attaining higher Sharingan power

Because of this, Tobirama was not convinced about the Uchiha Clan being peaceful people. On top of Hashirama's policy of having them becoming Konoha's police. The tendency to act in violence is high for them and therefore are not trustworthy people.

This is even up to the point of the Danzo conundrum. It was a self-fulfilling prophesy for both sides. To anticipate the uprising of the Uchiha, they must strike first to not be obliterated. To ensure the safety of the Uchiha, they must strike so that their clan was not destined to be destroyed.
