Tsunade invented a raiton based technique ( Ranshinsho ) that messes up the whole nervous system, so I doubt she can get paralyzed by any of the known raiton techniques.

That's an interesting thought. I don't think we'll be able to prove one way or the other for sure though.

Just because she invented a technique that does that that doesn't grant her immunity to paralysis.

Can she do it while paralyzed? Or is it a preemptive move?

Without being able to use Kamui for offense Kakashi would more then likely lose to either Tsunade or Mei in a 1v1 and now you add both of them together with Tsunade being able to boost Mei's huge Suitons even further as well as being able to boost her Yoton and Futton and Kakashi pretty much stands no chance.
