I'd say Itachi, his drive for vengeance against Itachi made him what he is now. Then VOTE 2 after learning the senju uchiha story and realizing the cycle of hatred which again indirectly made him into what he is and then making a decision to bring peace in his own way. Then I'd say Kaguya, as she came out of nowhere and nobody in the ninja world knew of her before that.

Itachi fight is valuable because he preparing for that fight the whole life but after the fight what sasuke did is exact opposite what itachi wanted... thus it became least important imo

On the other hand fight with naruto which change his life in positive manner

VotE2. Killing Itachi allowed Obito to tell Sasuke the truth about the Uchiha and set Sasuke on a path of vengeance against his real enemies. However, Sasuke was always destined to walk that path, and would inevitably have faced off against Naruto. VotE2 is where Sasuke finally broke the destiny of hatred between Indra and Ashura, and truly began to heal his emotional wounds.

Stopping Kaguya has to be hands down the least of his worries. He never cared that's how he barely even wanted to showcase his powers.

As for the most important, it is his fight with Itachi. He actually poured intelligent,creative ideas into that fight because his one goal in life was to kill Itachi. It meant so much to him.

I think it was the Itachi fight. That was his whole purpose for living until that point. Emotionally he was on the verge of collapsing and he did soon after learning the truth. That was his most important fight, because of the ramifications the fallout that ensued soon after created
