If Ashura had Uzumaki genes, then his children would also have Uzumaki genes. And their children would, and so on. Eventually, some of these people with Uzumaki genes would separate from the Senju to form the Uzumaki.
This is only a possibility that I am posing. I'm not saying this is what actually happened. But going by your idea of their mother being Uzumaki, this is what makes sense. How else would the Uzumaki have been formed going by your theory?

-You understood that Kito Uzumaki is the mother of Ashura and Indra and the wife of Hagoromo.
-But you didn't get that Sojobo Uzumaki would be the ancestor of the Uzumaki clan in my theory.
-Sojobo is Hagoromo' s best friend , Sojobo is the first human to have received the teaching of ninshuu from the sage , in my theory.
-Sojobo Uzumaki would fall in love with a girl and have babies named Uzumaki , creating a big family cause he loves life , giving the Uzumaki clan.
That's how Sojobo's descendance would have chakra and would be called Uzumaki.
