1.) He already has Kishi's blessing as being the next best choice besides Naruto
2.) For only being 13, he is incredibly powerful. Not quite on Part 1 Naruto or Sasuke level, but he can use an A rank jutsu in Rasengan and is quite proficient in Kage Bunshin, another high rank jutsu. Logically, he could inherit Enma as his summon from his grandfather, he could possibly have all elements in the future like his Grandfather Hiruzen (he at least has Katon like all Sarutobi), and he knows that being strong means you have to push yourself and go beyond your limits.
3.) He has the Will of Fire and realizes what it means to protect something you love, as demonstrated when he took on a path of Pain by himself. He didn't abandon his sensei and he fought valiantly, knowing what it means to have a village to look after and reach his goal of becoming like big brother Naruto. He'd have the support of everyone because of this, while conversely, Sasuke won't have the support of anyone.
4.) He's Naruto's student, which is a huge advantage. He can learn the true ways of peace from Naruto that Naruto learned from Jiraiya, Nagato, Itachi, and everybody else who's helped him become the ninja and man he is today. In imparting these to Konohamaru, Naruto would be fulfilling his role in handing down the Will of Fire like what Shikamaru said about teaching the new generations after Asuma's death.
5.) He's the grandson of Hiruzen Sarutobi, the God of Shinobi. Nothing is out of his reach. 'Nuff said.
So, Konohamaru is next in line after Naruto.
