First sorry if this title was misleading, this is only a speculation/hopeful thread. I have no information on who will actually be cast in the movie because it's becoming more and more likely this movie won't happen unfortunately.
Anyway. In a Deadpool movie I think we need a few core characters: Deadpool, Al, Weasel, T-Ray, Patch, Siryn, Killbrew and depending on if they cover the good vs evil battle Typhoid Mary.
First Deadpool:
I think Miles Teller would be a good fit. He's 6'0'' and as I mentioned in another thread the Marvel Encyclopedia has Deadpool at 6'2'' so not a far stretch.
For those who don't know him by name this is him, if you don't like his looks, Deadpool wears a mask.
He was in Project X, 21 and over, That Awkward Moment.
Why I think Miles would be a good choice is he has the tall thin build that should be associated with Deadpool rather than the bulky body. He doesn't have a too high pitched voice as I see a lot of people complain about the voice for Deadpool voice over things, he has really good comic timing(or at least what I've seen from him) and most importantly when playing someone nicknamed Merc with the mouth you need to be good at dialogue.

Weasel: My choice for Weasel has to be Jay Baruchel. Ever since I saw this fan pic it just makes absolute sense.
Here's another pic if you need a comparison.
I'm a huge fan of Jay's, I think he gets typecasted by fans as the nerdy guy, which would work for this role, but he has a lot to offer. now Jay may be taller than what they have Weasel listed as(5'7'' Jay being 6'2'') and I can see how some may have a problem with Weasel being taller than Deadpool but fear not because that's what editing is for.
Al: There's quite a few choices here for this one, in the place I found the Jay=Weasel pic they have Glenn Close as their Al
I think the obvious choice would have been Bea Arthur unfortunately she passed right before the release of Wolverine:Origins.
Another good possibility I think, would be Cloris Leachman. She's been in tons of stuff but you may remember her most from Malcolm in the Middle. I think that attitude without the accent would be a perfect fit for Al.
Lastly after seeing the previews for Tammy, I think Susan Sarandon could be a decent Al.
T-Ray: This is kind of a tough call because, and yes I am generalizing, bulkier actors aren't generally good actors if you know what I mean. There's the exceptions like Michael Clark Duncan, Matt Willig(whose shown to be a pretty good comedic actor), Eli Roth. This leads me to my pick for T-Ray or at least a candidate, Eli Roth, the bear jew from Inglorious Basterds. He was also in The Man with Iron Fists so he has a bit of a history with fighting movie roles. Matt Willig would be the better choice for the height difference. Then again there's always the choice Guardians took and hire a WWF(E) wrestler. Brock Lesnar comes to mind as someone whose physique matches pretty well.

Patch: Honestly I haven't considered him too much I figured Danny Devito or Peter Dinklage
Siryin: I think the obvious(Hollywood) choice would be Emma Stone. I'm fine with that, I've seen others say Isla Fisher(the girl from Wedding Crashers and Now You See Me). Whoever it is all I ask is they make her learn an Irish accent to do.

I was gonna mention Cable but I was figuring it being Deadpool after X Force so the Cable interaction would be minimal, maybe like an appearance at the end to hint towards an X Force movie or a Deadpool and Cable movie. If I picked Cable, I know it might be cliched but, I'd go with Ron Perlman. Butler would be a good choice. I've heard people say R. Lee Ermery(sp.) from Full Metal Jacket. Ron and Emery would need to bulk up or rely on editing like Hellboy. i thought this was a pretty good pic
