Nothing In Yamato's sleeve can hurt Tsunade...his jutsus are often used to either restrain targets or crush them, Tsunade can deal with both cases with her immense and superior strength.
PS: Yamato is only hyped because he can partially "control" the kyuubi, he's not that powerful battle-wise.
Exactly his only chance of winning would be taking the restrain method which she will have trouble breaking when even 4Tail kyuubi naruto had trouble loose of yamato's mass mukoton. Yamato has been shown to be able to suppress chakra and control it through mukoton similar to hashirama. Once he restrains tsunade he will restrain her with mass mukoton and suppress her chakra to stop her regeneration.
I believe that was a special method for only restraining the Kyuubi, Yamamto only used it when he's up against the kyuubi, which, pretty much, confirms it.
Also, Tsunade's strength can help her against the trees and roots, let alone summoning that acid-spitting slug (Forgot the name.).
He actually broke out of the mukuton (4tails), so the wood did nothing but buy a couple of seconds, it was not successful in restraining the 4 tails. Plus it was not strong enough to restrain a weaken kisame.. Tsunade is stronger then both ninjas! Plus Tsunades strength is not chakra enhanced, so no mukuton can keep her in check.
Tsunade is actually pretty quick on her feet, so Yamato would have to use a series of combos to defeat her. I don't see him winning, i have her winning more times then losing.

First off don't compare that weak ass mukoton yamato used on kisame, that shit was petty compared to other restrains he has shown. Not too mention even if tsunade manages to break it he can simply keep adding more mukoton. I know her strength isn't chakra enhanced but her regeneration is, that being said yamato can suppress her chakra if needed. And I know 4Tails broke out of mukoton regardless he was still restrained for a few mins. how exactly is tsunade touching yamato?
