Intel: Basic knowledge - Yugito knows Tsunade is a renowned Sannin and medical ninja. Tsunade knows Yugito is a jinchuuriki from the Hidden Cloud.

Conditions: Yugito is not edo
Restrictions: Tailed Beast Bomb if Yugito can use it but I doubt she can.

The fact that Yuugito could get injured by the likes of Kakuzu and Hidan implies that Kakuzu has the firepower to break through her defenses and spill her blood. Kakuzu's most destructive attack in terms of damage per unit area is raiton gian, which is hyped to pierce through rock but EP2 Sakura can destroy large boulders and war arc Byakugou Tsunade is leagues above this level of damage, so her punches, which can damage V1.5 susanoo, should be doing serious damage to Yuugito if not outright killing her due to possesses stronger attacks than the Zombie duo. Yuugito's long ranged attacks did not faze Hidan, so with regeneration, Tsunade should tank them just fine. Yuugito doesn't have any CQC feats and can't afford to get hit by Tsunade whereas Tsunade doesn't have to worry about getting hit so it will allow Tsunade to have the offensive edge in CQC which ultimately results in Yuugito getting killed.

I didn't even talk about Katsuyu yet but Tsunade has this in the bag with low-mid difficulty.

Tsunade as the Hokage and one of the Sanin automatically has knowledge on how to deal with jinchurikis, and this combined with her immense combat prowess.. uhh yeaaa she takes it.
