Your saddest Moment in Anime/Manga

Clannad: After Story. There are alot of sad scenes in it. But, I would probably go with either,

when Usiho lost her toy and then cried in Tomoya's arms. Or when Nagisa died.

One Piece is actually full of sad moments, mostly the flashback stories of characters. To name some:

Nami's flashback
Chopper's flashback
Robin's flashback
Ace's, Luffy's and Sabo's flashback
Montblanc Noland's and Calgara's flashback
Merry's death
Ace's and Whitebeard's deaths

That's just the few I consider the saddest. For a goofy shonen it has surprisingly a lot of those. Of course most people brush off One Piece as kids anime without knowing just how wrong they are.

Sad moments from other animes:

LOGH - Kircheis, Yang's and Reinhard's deaths
Death Note - Light's and Misa's deaths, basically the ending
Tengen Toppa Gurren Lagann - Kamina's death
Towards the Terra - Matsuka's and Sam's deaths
Code Geass - Lelouch's death
Naruto - Jiraiya's and Asuma's deaths
Dragon Ball - Krillin's death (first one, lmao), little indians father's death, Goku's death, bunch of more that I don't remember
Hellsing - Pip's death
Our Happy Hours, Our Happy Time - whole manga was one big tear jerker

Naruto: Jiraya's death, Minato's death, kushina's death... All in all, it's kind of a verry sad story ... everybody ends up dying around him
Bleach: his mothers death
One piece: that "brother"'s death (i'm not quite a fan, so...)
Fullmetal Alchemist: all of it...

Angel Beats - Yui & Hinata moment, and when Kanade disappears.

Mahou Shoujo Madoka Magica - When Madoka wishes for everything to change and save all the previous magical girls at the cost of her existence.

Ano Hi Mita Hana no Namae wo Bokutachi wa Mada Shiranai - When Menma disappears.

Steins; Gate - When Okabe goes through the time loop to try and save Mayuri, Also when he tries to save Kurisu.

Tengen Toppa Gurren Lagann - When Nia disappears.

Magico (IWAMOTO Naoki) - The price Emma & Shion, as well as their friends, have to pay at the ending.

Sora no Otoshimono - Sohara & Hiyori die.
