She used the regeneration jutsu right from the beginning. In one scan you can see that she has cuts all over her body. So if she wouldn't use her regeneration, the cuts would get bigger and bigger and eventually "rip to shreds".
But she did use the justsu, that's why the cuts didn't get big enough to rip her to shreds.

Yes, Tsunade was granted the body of the sage which makes her abnormally durable, albeit not as durable as the rakage but still notable durable. Note how she needs not wear gloves to protect her hands when she uses her strength.

A lot of people seemingly believe Byakugou healed those wounds. But if Byakugou had been active you would have seen marks over her entire body like here

So we know that the healing jutsu used was in fact not Byakugou. People are mistaking the markings of Creation Rebirth with Byakugou. Creation Rebirth however doesn't continually heal your wounds. Because she had to release chakra from the seal after she had already kicked Madara we can know that regeneration is not how she survived that jutsu as the seal was completely intact before landing.
