Why was orochimaru sick

I don't understand why orochimaru was so sick to the point he couldn't master Sage mode. Explain

Yeah but wouldn't Sasuke's decay after 3 years? If the whole thing that Sasuke's body was strong enough to handle the body transfer jutsu as well?

He probably gave Sasuke the curse seal to see if his body could handle nature energy and with Sasuke his body and the Sharingan he may had a plan if hebwould sucxeed in getting Sasuke's body.

Oro even had a hypotheses about the Rinnegan
He experimented with sharingans and Hashi dna.
He even had that Shin guy and clones who's genetic make up
didnt reject implants.

Its likely that Oro had plans with Sasuke's body and that he would be able to do more with it than using it as a 3 year temporary vessel.
