Well sakura did once take zetsu dna to test and she found out it was yamatos and that it uses a weaker ver of hashirama wood .

But things like you said needs human test objects .tsunade and sakura are not the type to use scarifies.

Tsunade and Sakura dont have that mind set to experiment with cells and implantation.

They would need to do human trials and Tsunade and Sakura would never do something like that.
