from what we've seen of everyone, I think its a pretty safe bet to say that Tsunade is the most powerful kunoichi around the block. We've never truly seen Mito or Kushina fight, so Id say Tsunade is the strongest shortly followed by Mei

She was the Kurama's first Jinchuuriki and she sealed Kurama in his entirety within herself by herself....Not to mention she had control over Kurama's Chakra...

That is more than enough evidence to say she was/is the Strongest Female Ninja,IMO.
