And the Senju clan is renowned for their large chakra reservoirs, and Tsunade has demonstrated to have a larger Chakra Capacities Than Gaara, and any other of the 5 Kages for that matter (the current kages that is)

She did not Inherit it from Mito. She developed it by herself, as stated in the manga i believe.
She remodeled the structure of the seal after her grandmother's marking on her forehead!
