I dont think it was Kaguya, Hagoromo, Indra or Ashura but maybe Hago's brother?

Or it could have been some random person who saw how the sage's healing powers worked and created their own healing.

Looking back at Madara and Hashirama's past, there didnt seem to be a lot of medical ninja. But I am sure it existed before their time.
