So I think you all remember when Tobirama said "I've never seen a Kagutsuchi that big before" when he saw Sasuke use it.
Kagutsuchi is the ability to control the black flames of Amaterasu.

There's Kagami his underling, and Satsuna Uchiha who was opposed to Tobirama. He had the ANBU watching them and Satsuna disappeared, so could've been assassinated.

Madara and his bro are supposed to be the first (in a long time, at least) Uchiha to awaken MS, so it is someone after them.

Fugaku said it was a major thing for someone to have MS when talking to Itachi (?) and Jiraiya did not know about MS, so the thing is rare even within the clan.

This leaves out everyone . . . besides the brothers themselves. I think, therefore, that it was either Madara or his brother. Most likely Madara because of the similarity to Sasuke. Most likely his brother because of the lack of visual evidence after many fights with Hashirama - think: fire which can change forms vs wood!
