He has the power, the loyalty to the clan (most loyal period), and the mindset of a leader.
It's between him and Obito, because Itachi didn't prioritize the clan, Shisui wasn't powerful enough or notable other than his sharingan.
Madara and Izuna weren't notable in smarts or leadership and Izuna isn't stated to have a Susanoo. Both are rivaled in power by Sasuke and Itachi.
