In the recent chapter someone pointed out its appearance in the seal ritual done for Kaguya. It was also placed in Sakura's spot.
When the hands are slammed together, a rhombus shaped space appeared. Maybe its a coincidence, but hand aren't normally like that when you clasp them together.
We know that its an Uzumaki Sealing Jutsu....most likely designed to contain Kuruma.
Shizune explains that one most store chakra in a single spot over a long period of time to fill it up. So this seal is able to contain the chakra of even the stongest of tailed beast.
How?
Look at all Sakura accomplished.
She healed the injured people in the shinobi alliance all at once, via Katsuyu. (by herself and the Shinobi alliance was about 80,000 people)
She did the same thing again, but with Tsunades help this time, via a bigger Katsuyu.
Lost some chakra from the God Tree, through her Katsuyu pieces, caused her a faint.
Lol, and Katsuyu is fine.

And Travel interdimensionally 4 time,
that cause 1/50? of a SO6P Naruto clone to poof away.
So right there proves that a seal is capable of holding the full chakra of Kuruma and then some.
Which would explain Mito's seal, and also why this was possible.
Im think Mito applied what Naruto did to extract Kuruma's chakra to the method used to form the yinseal. She wasn't renown for her great sealing powers for nothing.
Also, as seen with Tsunade one could use the seal as a conduit to connect you to a beast or summoning. Another possible way Mito tapped into kuruma's powers, without being called a perfect Jink.
Tsunade is granted a form of clairvoyance, her sensory and visual skills are directly linked to katsuyu...
She can also use this through Sakura...or the slug she summoned, it wasn't really clear.
But what about its weird healing power, the power it has to cheat death.
That is the secret part?
I think it has a connection the the reaper.
Tsuande's powers are able to undo death, and cheat it. In exchange for lifeforce.
No matter what she just wont die, and the only time she came close to death was due to plot chakra exhaustion.
