Prime means when one is at their best or strongest, for most people this is when they were young, but in the naruto world it means at any point when they were strongest.

Prime madara= JJ madara

Prime obito= juubito

Prime kakashi= DMS kakashi.

The databook specifically stated that Tsunade reached a pivotal position during the Second Shinobi World War.
Tsunade also went into elaborate that Kabuto was more efficient then she was in her prime, which can be concluded as being the Second Shinobi World War.
