almost all of the super OP jutsu come at a price (that's why they are called forbidden jutsu/harms the user) except for of course EDO TENSEI, which in this case is called "forbidden" because it has the potential to end the world when used properly (the price is the moral dilemma but in kabuto/orochimaru's case that's not a problem, moral and oro/kabuto cannot exist in the same sentence).

Tsunade's about 50 - 60 and yet her true appearance looks to be of someone 100+ So, yes, it shortens your lifespan.

We are talking about the rebirth using the stored up chakra to heal immediately by releasing it not storing up of chakra and using it to summon or fight
