jutsu allows for Tsunade to force her body's physiology to allow rapid regeneration of all her cells which need work. The cells undergo Mitosis, rather than fast-repairing of cells already alive. Elementary stuff we already know.Anatomy 101: The body has trillions of living cells within oneself. Each mitosis is semi-conservative, as one cell replaces the father and completes two. Every cell does this to accommodate the loss of worn-out cells.
Anatomy 504 (still ongoing research): The main reason why people die (natural causes) is likely due to the telomeres within the cells, which are affected by Mitosis. Each Mitotic session splits the telomere ever-so-slightly. Once the telomeres are shortened enough, they cannot synthesize the DNA required to continue living and it shuts down. The telomeres can only be split so many times before they call it quits.
We know confirmation from Shizune that every time Tsunade uses Creation Rebirth jutsu she "risks" shortening her lifespan to a certain degree. Let's tally how many times Tsunade has used Creation Rebirth jutsu to our knowledge:

How much of a degree does Tsunade lose her lifespan? How many more times will she be forced to use this jutsu when the five Kage come to the rescue? Each major injury Tsunade succumbs to requires these cells to split their telomeres several hundred times over to hasten her recovery in such a manner. The loss of blood, skin, bone, and tissues of several organs is too heavy a loss.
Barring decapitation or compromise of the heart, it's doubtful that Tsunade will die via combat, but there's no doubt that her body will be damned worn for the constant Mitotic reproduction of cells. She will die later, when all things are said and done.
